TEFOR-AMAGEN (UMS 3504 CNRS / UMS 1374 INRA) is located on 2 sites: Gif-sur-Yvette and Jouy-en-Josas. Its aim is to promote the utilization of aquatic animal models. TEFOR-AMAGEN has developed two models of teleost fishes: medaka (Oryzias latipes) and zebrafish (Danio rerio). The issues of modern biology involve the implementation of techniques allowing comparative analysis of living organisms.
Among vertebrates, the fish model displays easy high density breeding (short generation time), favourable biological peculiarities (accessibility and transparency of the eggs, genome sequenced for both species used on the platform), original technological approaches (confocal and two-photon microscopy), as well as a wide accessibility to fully transparent larvae usable up to 5 days after fertilization.
Transgenesis is an essential technique for studies in vivo. In a research and development process improving the transgenesis techniques in both species, TEFOR-AMAGEN will provide customized transgenic lines to European research teams.
Since its creation in 2009, TEFOR-AMAGEN has conducted research projects in the area of high specificity nucleases to develop targeted transgenesis and genome editing methods. In the frame of TEFOR, an infrastructure laureate of Future Investments in 2012 and focusing on non-mammalian animal models, TEFOR-AMAGEN has increased its capacity to produce zebrafish lines. The platform now proposes techniques to generate zebrafish lines via genome editing and KO (Knock-Out) models.
Aquatic animal models
- To develop and promote the utilization of aquatic animal models by scientific researchers : two models of teleost fishes (medaka, Oryzias latipes and zebrafish, Danio rerio).
- To improve breeding techniques of these aquatic species and to make the results available to the international community.
- To maintain an in-depth expertise in the transgenesis of aquatic species
- To develop and implement new techniques of:
• transgenesis in aquatic species
• characterization and detection of transgenes in aquatic species
- To develop efficient transgenesis vectors available to the scientific community by using the latest advances in that field
- To help research units to implement medium and high throughput phenotyping protocols.

A 57-hour course (45 hours training courses + 12 hours practical work) for research scientists and engineers managing a direct scientific authority in experimentation on animals. This training course includes the program for staff participating to experimentation on animals, such as technicians. That multi-site CNRS training has been implemented thanks to the collaboration of three partner centers of CELPHEDIA: TEFOR-AMAGEN, CRB Xenopus and CE2F-PRIM.

⑥ Zoology, comparative anatomy, general physiology, ethology, reproduction

TEFOR-AMAGEN is involved in several ambitious projects connecting academic laboratories and private companies
TEFOR-AMAGEN has developed a transgenesis platform entirely dedicated to the production of models for scientific research, either public or private, French or European.
- Development of Knock out (KO) technologies targeted via TALE nucleases, CRISPR and meganucleases
- Development of transgenesis and FO phenotyping via TALE nucleases (in the frame of TEFOR infrastructure)
- A set-up project of a zebrafish animal facility displaying a high health status level: SPF (Specific Pathogen Free/EOPS).
- In the frame of its involvement in TEFOR infrastructure and in cooperation with their imaging service as well as with academic partners specialized in zebrafish phenotyping, TEFOR-AMAGEN contributes to the development of new phenotyping tools in confocal imaging of the whole body with cellular resolution. The aim is to provide the scientific community with the required tools to perform expression profile screening of regulatory sequences directing transcription and to promote 3D imaging of transgenic lines.
